Functionsaskeymediatorinapoptosisandinflammation.Promotescaspase-mediatedapoptosisinvolvingpredominantlycaspase-8andalsocaspase-9inaprobablecelltype-specificmanner.Involvedinactivationofthemitochondrialapoptoticpathway,promotescaspase-8-dependentproteolyticmaturationofBIDindependentlyofFADDincertaincelltypesandalsomediatesmitochondrialtranslocationofBAXandactivatesBAX-dependentapoptosiscoupledtoactivationofcaspase-9,-2and-3.Involvedinmacrophagepyroptosis,acaspase-1-dependentinflammatoryformofcelldeathandisthemajorconstituentoftheASCpyroptosomewhichformsuponpotassiumdepletionandrapidlyrecruitsandactivatescaspase-1.Ininnateimmuneresponsebelievedtoactasanintegraladapterintheassemblyoftheinflammasomewhichactivatescaspase-1leadingtoprocessingandsecretionofproinflammatorycytokines.ThefunctionasactivatingadapterindifferenttypesofinflammasomesismediatedbythepyrinandCARDdomainsandtheirhomotypicinteractions.Requiredforrecruitmentofcaspase-1toinflammasomescontainingcertainpatternrecognitionreceptors,suchasNLRP2,NLRP3,AIM2andprobablyIFI16.IntheNLRP1andNLRC4inflammasomesseemsnotberequiredbutfacilitatestheprocessingofprocaspase-1.IncooperationwithNOD2involvedinaninflammasomeactivatedbybacterialmuramyldipeptideleadingtocaspase-1activation.MaybeinvolvedinDDX58-triggeredproinflammatoryresponsesandinflammasomeactivation.Isoform2mayhavearegulatingeffectonthefunctionasinflammasomeadapter.Isoform3seemstoinhibitinflammasome-mediatedmaturationofinterleukin-1beta.IncollaborationwithAIM2whichdetectscytosolicdouble-strandedDNAmayalsobeinvolvedinacaspase-1-independentcelldeaththatinvolvescaspase-8.InadaptiveimmunitymaybeinvolvedinmaturationofdendriticcellstostimulateT-cellimmunityandincytoskeletalrearrangementscoupledtochemotaxisandantigenuptakemaybeinvolvedinpost-transcriptionalregulationoftheguaninenucleotideexchangefactorDOCK2;thelatterfunctionisproposedtoinvolvethenuclearform.Alsoinvolvedintranscriptionalactivationofcytokinesandchemokinesindependentoftheinflammasome;thisfunctionmayinvolveAP-1,NF-kappa-B,MAPKandcaspase-8signalingpathways.ForregulationofNF-kappa-Bactivatingandinhibitingfunctionshavebeenreported.ModulatesNF-kappa-BinductionattheleveloftheIKKcomplexbyinhibitingkinaseactivityofCHUKandIKBK.ProposedtocompetewithRIPK2forassociationwithCASP1therebydown-regulatingCASP1-mediatedRIPK2-dependentNF-kappa-Bactivationandactivatinginterleukin-1betaprocessing.
